Abstract

An overarching goal of current research in our lab is to assess the impact of mitochondrial transfer between cancer cells and their non-cancerous neighbors, focusing on epigenetic and metabolic impacts. Specifically, we are using SW1353 cells, a chondrosarcoma cell line, carrying a mutation in the IDH2 gene which disrupts the TCA cycle and generates an important oncometabolite, D-2-hydroxyglutarate (D2-HG). We will test the impact of transfer of mitochondria from SW1353 to non-cancerous recipients, as well as return trafficking of mitochondria from the non-cancer cell to SW1353’s. A challenge we will face in these experiments arises from the fact that the recipient cells, either SW1353 or non-cancer cells, will contain 100-1,000s of their resident mitochondria. To enhance our ability to detect the impact of mitochondrial transfer, we are developing mitochondria-free cell lines, these are designated as rho0 cell lines. Techniques for targeting mitochondrial for depletion as well as our preliminary results will be presented.
